三磷酸腺苷结合盒转运体(ATP-binding cassette transporters, ABC)是一组跨膜蛋白,与ATP结合在胞膜和内质网,过氧化物酶体,线粒体等细胞器膜上,介导氨基酸、脂质、脂多糖、无机离子、多肽-糖类、各种药物等多种分子的耗能转运。目前已知的人类ABC基因包括A、B、C、D、E、F、G7个家族,共48个成员,ABC转运体的变异可以相应引起多种疾病,比如囊性纤维病,脑白质肾上腺萎缩症等。近年来,人们发现一些ABC家族成员与脂质代谢有关,并参与胆固醇结石等疾病的发生、发展过程。
ABCA1基因位于9q31,1,约149kb,其编码一种含有2261个氨基酸的跨膜蛋白,主要参与胆固醇逆转运,即HDL将外周组织过多的胆固醇转运到肝脏的代谢过程。ABCA1将细胞内胆固醇和磷脂转运至细胞表面的HDL前体aooA1,该蛋白负责胆固醇逆转运的第一步也是主要的限速步骤,因此在胆固醇和血浆脂代谢中起着重要的作用。ABCG5和ABCG8是近年被发现的参与植物固醇和胆固醇吸收的基因,与β-谷固醇血症有关,这对基因定位在染色体2p21上,仅相距374个氨基酸,两者都含有13个外显子和12个内含子,分别编码sterolin-1和sterolin-2蛋白,它们在肝脏的肝细胞、胆管细胞、小肠上皮细胞以及胆囊上皮细胞中表达。
